Calbindin 1, also known as calbindin D28K, is a member of a family of calcium binding proteins. This protein family also includes calmodulin, calbindin 2, S-100, and troponin C. Calbindin 1 is a 261-amino acid protein with 6 EF-hand domains, 4 of which are active calcium-binding domains. Expressed in brain, ovary, uterus, testis, pancreas, liver, kidney and intestine, calbindin 1 acts as a calcium-buffering agent and alters the activity of the plasma membrane ATPase. In neuronal cells, calbindin 1 modulates calcium channel activity, calcium transients and intrinsic neuronal firing activity. This antibody recognizes a protein of 54 kDa, which is identified as Alpha-1 antitrypsin (AAT). AAT is secreted and is a serine protease inhibitor whose targets include elastase, plasmin, thrombin, trypsin, chymotrypsin, and plasminogen activator. This antibody cocktail recognizes three melanoma-specific proteins, which include MART-1, Tyrosinase and gp100. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Function of gp100 is not known but it is reported to be a useful marker for melanocytes and melanomas. This cocktail of three markers is designed for extremely sensitive labeling of formalin-fixed, paraffin-embedded melanomas and other tumors showing melanocytic differentiation.

Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer. It acts as a carrier protein for Factor VIII and promotes platelet adhesion and aggregation. vWF undergoes a variety of posttranslational modifications that influence the affinity and availability for Factor VIII, including cleavage of the propeptide and formation of N-terminal disulfide bonds. This antibody helps to establish the endothelial nature of some lesions of disputed histogenesis, e.g. Kaposi's sarcoma and cardiac myxoma. The wild-type anaplastic lymphoma kinase (ALK) protein is a 200 kDa transmembrane receptor tyrosine kinase. Its expression is restricted to a few scattered cells in the nervous system (some glial cells and neurons, and a few endothelial cells and pericytes). ALK has been found to be rearranged, mutated, or amplified in a series of tumors. Spectrin is an actin binding protein that is a major component of the plasma membrane skeleton. Spectrins function as membrane organizers and stabilizers by forming dimers, tetramers and higher polymers. Vertebrate spectrins have two alpha-subunits (alpha-I/alpha-II) four beta-subunits (beta-I-beta-IV) and a beta-H subunit creating diversity and specialization of function. Spectrin and spectrin are present in erythrocytes, whereas spectrin II (also designated fodrin ) and spectrin I (also designated fodrin ) are present in other somatic cells. The LMO2 protein has a central and crucial role in hematopoietic development and is highly conserved. It has a particular function in normal and lymphatic endothelial cells involving the regulation of angiogenesis and lymph-angiogenesis. Immunohistochemical studies have also demonstrated expression of LMO2 in both normal germinal center B-cells and germinal center-derived B-cell lymphomas, including follicular lymphoma and diffuse large B-cell lymphoma. The use of anti-LMO2 is valuable as a tool in the identification of lymphomas of B-cell origin. Recognizes a protein of 55 kDa, which is identified as GLUT-1. Glucose transporters are integral membrane glycoproteins involved in transporting glucose into most cells. This MAb recognizes a protein of 36 kDa, identified as ICOS-L. It is a ligand for the ICOS receptor that initiates T and B cell proliferation and cytokine secretion. ICOS-L interactions play an essential role in T cell-dependent B cell activation in peripheral lymphoid organs such as spleen and lymph nodes. ICOS-L protein is present in myeloid leukocytes, and by Northern blot there are 2. 4, 3. 0, and 7. 0 kb transcripts in brain, heart, kidney, and liver, with lower expression in colon and thymus, and a 1. 1 kb transcript in leukocytes. Recognizes a protein of 30 kDa which is identified as Prohibitin, an evolutionarily conserved protein with homologues found in yeast to man. It is located in the inner membrane of mitochondria. Although prohibitin mRNA and protein expression occurs throughout the cell cycle, maximum levels are detected during the G1/S phase transition and minimum levels are seen in S phase and the G2/mitosis boundary. Prohibitin is located exclusively in the mitochondria with the highest concentration on the inner membrane. Prohibitin is an ideal mitochondrial marker. It shows antiproliferative activity and has been proposed to play a role in normal cell cycle regulation, replicative senescence, cellular immortalization, and tumor suppression.
